«Acting on gut microbiota in metabolic diseases ; for who, what and when ? »
As Director of the Inserm/Sorbonne University research team at Sorbonne University (Faculty of Medicine), Paris, France, I am coordinating efforts to understand the complex pathophysiology of human obesity and related disorders, using large scale and translational approaches. I have been involved in genetic and functional genomics aspects of human obesity, and contributed to the identification of monogenic forms of obesity (Leptin receptor and MC4R mutations). Today patients suffering from these diseases benefit from new targeted treatments. Using functional approaches in commoner forms of obesity, I showed notably that inflammatory and remodeling genes in different metabolic tissues are modulated by weight variation in parallel to immune cell infiltration changes. Deeper insight into cellular mechanisms were undertaken showing the contribution of profibrotic progenitors in adipose tissue. My research team is currently exploring the link between environment changes, immune systemic changes and functional modifications in the adipose tissue using different mouse and human models. The gut microbiota is of evidence a key actor of these links and explorations of this organ is currently deeply undertaken. We demonstrated the importance of low gut microbiota diversity (as part of dysbiosis) in the severity of moderate to severe obesity and the possibility to (at least partially) rescue gut dysbiosis using different intervention (dietary or gastric surgery). Further insight into the mechanistic understanding of the relationships between environment changes, gut microbiota dysbiosis and altered tissue metabolism and inflammation are now undertaken.
